Overview

Argireline (Acetyl Hexapeptide-3, now also called Acetyl Hexapeptide-8 per INCI nomenclature) is one of the most commercially successful cosmetic peptides globally. It was designed to mimic the N-terminus of SNAP-25, a SNARE complex protein involved in vesicle docking and neurotransmitter release at the neuromuscular junction. By competing with SNAP-25 for SNARE complex formation, Argireline reduces acetylcholine release at the NMJ, temporarily relaxing facial muscles and reducing the depth of dynamic expression lines. It is nicknamed "topical Botox" though the mechanism and penetration are fundamentally different from botulinum toxin.
